MICROFLUIDIC DEVICES FOR MEMBRANE PROTEIN CRYSTALLIZATION AND STRUCTURE DETERMINATION

Abstract

Structure determination of membrane proteins is crucial to design targeted drug molecules for therapeutic interventions of diseases caused by membrane protein dysfunctions. The structures of membrane proteins can be obtained in high resolution using X-ray crystallography. The limitation of low sample availability is hampering the advancement of the field which can be mitigated by using microfluidic devices with very low sample requirement to obtain optimized crystallization conditions as well as optimized sample delivery means. Here we report a microfluidic system which is capable of screening hundreds of crystallization conditions and subsequently be used as a fixed target sample delivery device for screening in X-ray crystallography.
