N2 - Statement of Purpose / Introduction: Mammalian tissue development is a multifaceted process of self-organization that emerges from gene regulatory networks of differentiating stem cells and their progenies. Stem cell derived organoids have shown high potentials to decode fundamental aspects of development while providing a platform for disease modeling and regenerative therapies. Current approaches to generate such cellular systems rely on recapitulating the developmental stages by supplementing growth factors or combining subset of cells. We propose a novel strategy to engineer selfvascularizing organoids via genetic engineering of developmental processes.
